Vitamin D receptors (VDR) are present in Leydig cells and are required for normal testosterone synthesis. Observational studies consistently show that men with lower 25(OH)D have lower testosterone levels. Multiple RCTs in deficient men show that vitamin D3 supplementation (2,000–5,000 IU/day) increases testosterone by 20–25%. The effect is most pronounced in men with baseline 25(OH)D below 20 ng/mL. Supplementation in replete men shows smaller or negligible effects.

Permitted in food supplements at up to 100 μg (4,000 IU) per daily serving under EFSA guidance. Upper intake level 100 μg/day.

United States · FDA

Dietary supplement ingredient under DSHEA. Tolerable upper intake level: 4,000 IU/day (NIH), though many clinicians use higher supervised doses.
